If you complete an area without unlocking all heroes in that area you can continue to unlock them even in the following areas. You can never get stuck with not being able to unlock a certain hero.
In case the new area provides new heroes too, a quest chooses between the unlockable heroes randomly.
Just keep completing quests and you will eventually unlock all heroes.
